Metafolin is the calcium salt of (6S)-5-methyl-5,6,7,8-tetrahydrofolate (CAS 151533-22-1; C20H23CaN7O6; MW 497.52), a reduced, coenzyme-form folate supplied as a single defined (6S) isomer. It is produced synthetically and isolated as the calcium salt to USP grade in powder form. In contrast to fully oxidized folic acid, its pteridine ring is reduced to the tetrahydro state and bears a 5-methyl substituent; and unlike the racemic calcium folinate it is delivered as one stereochemically pure (6S) isomer rather than a (6RS) mixture. Its calcium counter-ion distinguishes it from the glucosamine-salted L-methylfolate, and its molecular weight sits above folic acid owing to the added calcium and the reduced ring. We manufacture it in capsules, tablets, softgels, powders and gummies.
